When flying a B738 the pilot voice on FSX says for example, Orbit 5454 with MIKE!

What does MIKE actually mean, sometimes they also say with ECHO and various other word.

What do they all mean?

MIKE will mean the current airport ATIS (Automatic Terminal Information Service)

Each time it is updated throughout the day, it is assigned a different letter.

Pretty much all it is. Smile
